Route Description
  1. Southbound bus starts at Orient, Cross Sound Ferry Terminal.
  2. Travels west along Main Road, or New York State Route 25, through Orient and East Marion until turning south onto Greenport Main Street.
  3. Makes stops through downtown Greenport, including the Greenport Long Island Rail Road station before resuming westbound on Main Street, or New York State Route 25.
  4. Westbound bus makes stops at vineyards, breweries, cultural sites, and shopping centers through Southold, Cutchogue, Mattituck, Laurel, Jamesport, and Aquebogue with stops near the Southold and Mattituck Long Island Rail Road stations.
  5. After crossing Cross River Drive, New York State Route 25 turns into Riverhead East Main Street heading southbound with stops throughout downtown Riverhead before arriving at the Riverhead Long Island Rail Road station via Griffing Avenue, Court Street, and Cedar Street.
  6. Southbound bus heads south on Cedar Avenue to head west on Court Street to go east on Nugent Drive, or County Route 94, via Cedar Drive Spur, or County Route 94A.
  7. Stops at the Suffolk County Center via Center Drive South and reconnects with Nugent Drive, or County Route 94, to head towards the traffic circle and merge onto Flanders Road, or New York State Route 24.
  8. Proceeds east and makes stops near shopping centers and parks as the bus travels towards Hampton Bays.
  9. Reaches Montauk Highway, or County Route 80, and turns to travel east making stops in downtown Hampton Bays.
  10. After crossing Shinnecock Canal, southbound bus heads north on North Road, or County Route 39, until reaching the end of the road to loop back around North Road once again going south and turning east to merge onto Sunrise Highway, or New York State Route 27.
  11. New York State Route 27 turns into County Route 39 and southbound bus enters Southampton On-Demand zone.
  12. Turns south on North Sea Road and approaches downtown Southampton via Windmill Lane Nugent Street which turns into Hampton Road.
  13. Heads east on Meeting House Lane via Little Plains Road and continues north on Old Town Road.
  14. Proceeds eastbound on Montauk Highway, or New York State Route 27, via Hampton Road to make stops in Watermill and Bridgehampton.
  15. Turns north on County Route 79 or Bridgehampton-Sag Harbor Turnpike, with stops including East End Children's Museum, South Fork Natural History Museum, and Nature Center on the way to Sag Harbor.
  16. Makes stops along Main Street and Bay Street in Sag Harbor before continuing southeast on Hampton Street, or New York State Route 114, via Division Street.
  17. Continues southeast on Hampton Street which converts to East Hampton Sag Harbor Turnpike to make stops near downtown East Hampton.
  18. Travels north on Toilsome Lane and terminates at the East Hampton Long Island Rail Road station on Rail Road Avenue via Gingerbread Lane and Lumber Lane.
